Robust and scalable measurement of neutralizing antibodies is critical for influenza surveillance, vaccine development, and outbreak preparedness. Although focus reduction assay (FRA) is widely used to quantify neutralizing antibodies against influenza viruses, its application is limited by labor-intensive immunostaining workflows that restrict throughput. These limitations are particularly challenging when large sample sets must be analyzed during seasonal epidemics or emerging outbreaks caused by high pathogenicity avian influenza viruses. We sought to develop a streamlined assay that preserves the accuracy of FRA while reducing complexity and enabling high-throughput implementation. By leveraging neuraminidase activity as a direct readout of infection, DirectView-FRA addresses these bottlenecks and expands the practical utility of FRA.
